Do I have immediately to pay 2 pool or skip the rest of my turn, or the First tradition effect only triggers when a turn is starting?

The effects of the card are only applied at the beginning of a turn.  Passing the vote does not end and restart your turn.

A Methuselah can choose to skip his or her entire turn. Burn this card when the controller has either skipped 3 turns or burned a total of 6 pool by this effect.
